A high quality foam dewar integrates grid box storage puck access directly adjacent to the clipping space, increasing ease of use and reducing the potential for contamination transferring grid boxes from pucks in other stations. A new clipping design makes the workflow easier, and a well designed lid balances ease of use and protection.&nbsp;Furthermore, our Clipping Station is compatible with both NanoSoft and Thermo Fisher clipping tools.</p><p>​</p><p>The station is housed in a high quality, closed-cell polyethylene foam, i.e. the Spearlab foam. The foam is highly durable and cools down and warms up/dries off quickly. The inside of the foam dewar is designed to match the intricacies of the workflow; there are 3 cutout positions in the foam, each of which holds a component firmly in place such that it doesn't slide around during use. One cutout holds the clipping hub, which is the part used for clipping. The other 2 are the appropriate size to hold storage pucks. This allows you to place a storage puck directly and securely in the station adjacent to the Hub, so that grid boxes can be accessed without needing to transfer them through the ambient environment from another dewar. Furthermore, the Station comes with a puck sized holder for clipping tools, such that up to 33 tools can be pre-loaded with c-clips and placed in the holder to be cooled down while being held reliably in place; the puck and tools are in a position that is easy to access and enables seamless clipping of up to 33 samples at one time&nbsp;with one puck-full of tools. A valuable option is also to use the MiTeGen cassette puck - this enables you to load a cassette at the same time and in the same station where grids are clipped.</p><p>​</p><p>The Hub used for clipping utilizes a novel mechanism for moving the guidance cylinder, used to center the clipping tool over the autogrid ring, into and out of position. Instead of the rotating brass cylinder, which is often difficult to turn and can bend tweezers, the seesaw like mechanism requires the user to push down on either side of the cantilever beam to move the guidance cylinder into the position for clipping or into the position where the autogrid ring is placed or where the clipped grid can be picked up. Once the system and clipping tools are cooled down, and a storage puck placed into its spot adjacent to the Hub, the grid box holding the grids to be clipped can be moved to a grid box pocket in the Hub for easy grid access for clipping, and replacement of the autogrid after clipping. &nbsp;Furthermore, The Hub has 4 "on-deck" positions for clipping tools where they can quickly be accessed for rapid-fire clipping. The shallow hole where the autogrid ring is placed for clipping has a dark colored surface to provide better contrast and easier tracking for placement of the autogrid ring and picking up of the clipped grid with autogrid tweezers. Furthermore, grooves are machined into the Hub to enable inspection of the clipped grid to ensure that it is well clipped and to roll the notch of cryoET, FIBSEM rings into the right orientation.</p><p>​</p><p>The lid for the station provides a thoughtful balance between usability and efficient separation of the clipping space from the ambient environment. The optimally designed opening in the lid provides the ideal amount of space to access the cantilever, the grid box pockets in the Hub and the 4 "on-deck" clipping tools, all without needing to rotate the lid, and while providing protection from ambient humidity. Knobs on the lid enable easy turning to access and transfer other grid boxes or clipping tools to the Hub, and a Lid insert can cover the entire opening so that the clipping space can be closed off from the ambient environment completely during cool down and as needed. Customer must ship their Linkam cartridge to NanoSoft for modification.</p><p>&nbsp;</p><p>The CryoCLEM cartridge adaptor enabling direct insertion of the Linkam cartridge (that holds unclipped grids or autogrids for cryo-fluorescence imaging) into the Leica shuttle (for use with the VCT500 system, for insertion into Zeiss FIBSEM) reduces the number of direct manual manipulations of the grid during sample transfers. The Linkam CMS196V³ stage used for cryoCLEM imaging along with the Leica VCT500 for FIB milling of vitrified grids in a Zeiss FIB-SEM enables powerful insights into cells through the utilization of cryoCLEM-based, targeted ion beam milling and subsequent tomography. However, there are multiple direct handling steps of the vitrified grid to transfer them between the Linkam cartridge and the Leica shuttle.</p><p>&nbsp;</p><p>Our Linkam cartridge adaptor removes the need for any direct grid handling once it is placed in the Linkam cartridge. The vitrified grid is placed normally into the Linkam cartridge for cryoCLEM imaging. Our new adaptor and clamp attach directly to the Leica VCT500 shuttle, and once fluorescence imaging is complete the entire cartridge can be slipped into our adaptor and held tightly with the custom clamp. This removes the tenuous manipulation steps of the fragile grid, after the valuable CLEM imaging. Furthermore, we modify your Linkam cartridge to ensure that you have full ion beam access to the grid for lamellae milling. The custom clamp that holds the Linkam cartridge is locked, and unlocked, from holding the cartridge in place through a simple, easy to activate screw in the back of the Leica shuttle.</p> by the ambient environment and by the expertise of the user.&nbsp; The grid is exposed to air during the transfer - users report that they observe variations in sample quality based on time of year, weather, location, etc., which suggests ambient properties are influencing the sample.&nbsp; Manually transferring the grid&nbsp;requires dexterity while simultaneously tending to multiple things&nbsp;(liquid nitrogen&nbsp;levels, ethane quality, etc.) – it’s stressful for experienced users, and for beginners trashed grids are the norm.</p><p>​</p><p>NanoSoft's Igloo aims to better control the&nbsp;manual transfer when preparing samples with the Thermo Fisher Vitrobot.&nbsp; A more enclosed environment provides protection from contaminating ambient&nbsp;humidity, while various system features transform the manual transfer process into a user friendly experience. The Igloo is beneficial for all users, but less experienced users in particular will be able to prepare viable grids sooner.</p><p>​</p><p>Igloo Cover:&nbsp;The styrofoam ring from the Vitrobot dewar aims to maintain an&nbsp;environment containing&nbsp;cold, dry nitrogen gas above the liquid nitrogen to protect vitrified&nbsp;grids during their transfer.&nbsp; However, the ring is&nbsp;open and susceptible to humidity in the external environment.&nbsp; Furthermore, the ring is cold and collects ice.&nbsp; As&nbsp;it is submerged into the liquid nitrogen with every plunge of the Vitrobot, ice particulate is deposited into the liquid nitrogen, which can contaminate&nbsp;grids already placed&nbsp;in&nbsp;the grid box.</p><p>​</p><p>The Igloo is a nearly enclosed cover that sits atop the Vitrobot dewar, with an opening large enough for comfortable grid button access and grid transfer.&nbsp; The volume enclosed by the Igloo fills with cold gaseous nitrogen, creating a dry space with improved protection versus the ambient&nbsp;environment.&nbsp; The Igloo is designed to fit comfortably over the floating styrofoam ring, minimizing the&nbsp;time that the styrofoam ring is exposed to ambient humidity and&nbsp;significantly&nbsp;reducing the potential for ice particulate contaminating the liquid nitrogen.&nbsp; The Igloo material is compatible with liquid nitrogen&nbsp;temperatures, allowing for easy refill with the Igloo on the dewar.&nbsp; Furthermore, the top surface of the Igloo remains comfortable to the touch, providing a convenient surface upon which the user can rest their hands for steady transfer.</p><p>​</p><p>The Cobra:&nbsp;Upon removing the tweezers from the Vitrobot dovetail, users must carefully hold the tweezers closed (to hold grid), keep the tips of the tweezers submerged in liquid ethane and simultaneously lift the dewar to transfer it away from the Vitrobot to transfer the sample to the grid box.&nbsp;</p><p>&nbsp;</p><p>The Cobra is integrated into the top surface of the Igloo and flips up to provide a slot into which tweezers can be placed.&nbsp; When resting in the slot, the tips of the tweezers remain submerged in liquid ethane.&nbsp; The Cobra slot is compatible with both NanoSoft's Cryo-Tweezers and other Vitrobot tweezers.&nbsp; The Cobra engages with magnets in the Plunge-Clamp of NanoSoft's Cryo-Tweezers, providing a more secure attachment. The Cobra provides freedom to the user to focus both hands on holding the dewar when moving it from the Vitrobot, the ability to fill the dewar with liquid nitrogen, etc.</p><p>​</p> user to manually place the tip of the nozzle from the ethane cylinder (e.g. pipette tip, bike pump needle, etc.) into the bottom corner of the brass ethane cup.&nbsp; The user handles the nozzle with one hand and must simultaneously control ethane flow rate with the other hand (e.g. with needle valve), while watching the liquid ethane level.&nbsp; The user will also often move the nozzle around the brass cup to avoid condensing ethane in a warmed position.</p><p>​</p><p>NanoSoft's custom designed ethane condenser tightly interfaces with the spider from the FEI/Thermo Fisher styrofoam dewar set.&nbsp; The user places the spider normally and inserts the condenser.&nbsp;&nbsp;The user does not need to handle an&nbsp;ethane nozzle&nbsp;and can instead focus solely on the liquid ethane level, using just&nbsp;one hand to control ethane flow rate, making the process much easier and ergonomic.&nbsp;Furthermore the condenser should reduce wasted ethane gas&nbsp;and the time required to Fisher autoloader-based microscopes, with its design optimized for the clipping workflow. A high quality foam dewar integrates grid box storage puck access directly adjacent to the clipping space, increasing ease of use and reducing the potential for contamination transferring grid boxes from pucks in other stations. A new clipping design makes the workflow easier, and a well designed lid balances ease of use and protection.&nbsp;Furthermore, our Clipping Station is compatible with both NanoSoft and Thermo Fisher clipping tools.</p><p>​</p><p>The station is housed in a high quality, closed-cell polyethylene foam, i.e. the Spearlab foam. The foam is highly durable and cools down and warms up/dries off quickly. The inside of the foam dewar is designed to match the intricacies of the workflow; there are 3 cutout positions in the foam, each of which holds a component firmly in place such that it doesn't slide around during use. One cutout holds the clipping hub, which is the part used for clipping. The other 2 are the appropriate size to hold storage pucks. This allows you to place a storage puck directly and securely in the station adjacent to the Hub, so that grid boxes can be accessed without needing to transfer them through the ambient environment from another dewar. Furthermore, the Station comes with a puck sized holder for clipping tools, such that up to 33 tools can be pre-loaded with c-clips and placed in the holder to be cooled down while being held reliably in place; the puck and tools are in a position that is easy to access and enables seamless clipping of up to 33 samples at one time&nbsp;with one puck-full of tools. A valuable option is also to use the MiTeGen cassette puck - this enables you to load a cassette at the same time and in the same station where grids are clipped.</p><p>​</p><p>The Hub used for clipping utilizes a novel mechanism for moving the guidance cylinder, used to center the clipping tool over the autogrid ring, into and out of position. Instead of the rotating brass cylinder, which is often difficult to turn and can bend tweezers, the seesaw like mechanism requires the user to push down on either side of the cantilever beam to move the guidance cylinder into the position for clipping or into the position where the autogrid ring is placed or where the clipped grid can be picked up. A high quality foam dewar integrates grid box storage puck access directly adjacent to the clipping space, increasing ease of use and reducing the potential for contamination transferring grid boxes from pucks in other stations. A new clipping design makes the workflow easier, and a well designed lid balances ease of use and protection.&nbsp;Furthermore, our Clipping Station is compatible with both NanoSoft and Thermo Fisher clipping tools.</p><p>​</p><p>The station is housed in a high quality, closed-cell polyethylene foam, i.e. the Spearlab foam. The foam is highly durable and cools down and warms up/dries off quickly. The inside of the foam dewar is designed to match the intricacies of the workflow; there are 3 cutout positions in the foam, each of which holds a component firmly in place such that it doesn't slide around during use. One cutout holds the clipping hub, which is the part used for clipping. The other 2 are the appropriate size to hold storage pucks. This allows you to place a storage puck directly and securely in the station adjacent to the Hub, so that grid boxes can be accessed without needing to transfer them through the ambient environment from another dewar. Furthermore, the Station comes with a puck sized holder for clipping tools, such that up to 33 tools can be pre-loaded with c-clips and placed in the holder to be cooled down while being held reliably in place; the puck and tools are in a position that is easy to access and enables seamless clipping of up to 33 samples at one time&nbsp;with one puck-full of tools. A valuable option is also to use the MiTeGen cassette puck - this enables you to load a cassette at the same time and in the same station where grids are clipped.</p><p>​</p><p>The Hub used for clipping utilizes a novel mechanism for moving the guidance cylinder, used to center the clipping tool over the autogrid ring, into and out of position. Instead of the rotating brass cylinder, which is often difficult to turn and can bend tweezers, the seesaw like mechanism requires the user to push down on either side of the cantilever beam to move the guidance cylinder into the position for clipping or into the position where the autogrid ring is placed or where the clipped grid can be picked up.
